建立了柱前衍生化液相色谱-串联质谱法测定血浆中的雌二醇。以雌二醇-d3为内标,采用C18色谱柱,血浆样品经液-液萃取、柱前衍生化处理后,采用ESI离子源,多反应离子监测(MRM),正离子扫描,进行测定。结果表明,雌二醇在50~10 000 pg/ml浓度范围内线性关系良好,方法回收率为94.0%~107.2%,RSD<10.0%。同时测定了雌性大鼠体内雌二醇基础值和给予雌二醇透皮贴剂后的血药浓度。结果显示本法灵敏度高、重现性好,适用于药动学研究。
A pre-column derivatization liquid chromatography-tandem mass spectrometry was established for the determination of estradiol in plasma. Using estradiol-d3 as internal standard and C18 column, the plasma samples were subjected to liquid-liquid extraction and pre-column derivatization. ESI source, multi-reactive ion monitoring (MRM) and positive ion scanning were used for the determination. The results showed that estradiol had a good linearity in the concentration range of 50 ~ 10 000 pg / ml. The recovery rate of the method was 94.0% -107.2% with RSD <10.0%. At the same time, the estradiol value in female rats and the plasma concentration of estradiol transdermal patch were measured. The results show that the method of high sensitivity, good reproducibility, suitable for pharmacokinetic studies.
