目的:探讨尼美舒利诱导人肝癌SMMC-7721细胞凋亡的作用及其机制。方法:以不同浓度的尼美舒利处理体外培养的人肝癌SMMC-7721细胞,MTT法检测细胞活力,流式细胞术检测细胞凋亡率,RT-PCR和/或Western blot法检测caspase-9和PARP的剪切情况及HSP70的表达水平;通过RNAi技术沉默HSP70基因表达,观察其对细胞凋亡的影响。结果:尼美舒利可抑制人肝癌SMMC-7721细胞生长,诱导细胞凋亡,导致caspase-9和PARP的剪切活化及HSP70的mRNA和蛋白表达下调;在人肝癌SMMC-7721细胞中转染靶向HSP70的siRNA可沉默HSP70表达、剪切活化caspase-9和PARP,进而显著促进细胞凋亡。结论:尼美舒利可诱导人肝癌SMMC7721细胞凋亡,这一作用可能与抑制HSP70表达有关。
Objective: To investigate the effect and mechanism of nimesulide on apoptosis in human hepatoma SMMC-7721 cells. Methods: Human hepatocellular carcinoma SMMC-7721 cells were treated with different concentrations of nimesulide. Cell viability was measured by MTT assay. Apoptosis rate was detected by flow cytometry. Caspase-9 expression was detected by RT-PCR and / or Western blot And the cleavage of PARP and the expression of HSP70. The expression of HSP70 was silenced by RNAi and the effect of HSP70 on apoptosis was observed. Results: Nimesulide could inhibit the growth of human hepatocellular carcinoma SMMC-7721 cells, induce apoptosis, induce the cleavage of caspase-9 and PARP, and down-regulate the expression of HSP70 mRNA and protein. The transfection of human mesangial SMMC-7721 cells SiRNA targeting HSP70 silences HSP70 expression, cleaves activation of caspase-9 and PARP, thereby significantly promoting apoptosis. Conclusion: Nimesulide can induce the apoptosis of human hepatoma SMMC7721 cells, which may be related to the inhibition of HSP70 expression.
