目的研究胰高血糖素样肽-2(GLP-2)对短肠大鼠残留小肠钠葡萄糖共同转运体(SGLT1)和二肽转运体(PEPT1)的mRNA表达影响。方法将75%小肠切除大鼠分为空白对照组(空白组)、生长激素对照组(GH组)和胰高血糖素样肽2组(GLP-2组),另设一组正常进食大鼠作空白组的对照组(正常组)。术后1d起进食标准大鼠饲料。术后6d取末端回肠黏膜,用逆转录多聚酶链反应方法半定量检测其SGLT1和PEPT1的mRNA表达情况。结果残留回肠SGLT1和PEPT1的mRNA表达,空白组显著高于正常组(P<0.05),空白组和GH组及GLP2组之间差异均无统计学意义(P>0.05)。结论GLP-2术后短期应用对短肠大鼠残留回肠SGLT1和PEPT1的mRNA表达可能无显著影响。
Objective To investigate the effects of GLP-2 on the mRNA expression of small intestinal sodium-glucose co-transporter (SGLT1) and dipeptide transporter (PEPT1) in short-term rats. Methods 75% of small intestine resected rats were divided into blank control group, GH control group and GLP-2 control group. Another group of normal rats The blank control group (normal group). One day after operation, the standard rat feed was taken. The terminal ileum mucosa were harvested 6 days after operation and the mRNA expression of SGLT1 and PEPT1 was semi-quantitatively detected by reverse transcription polymerase chain reaction. Results The mRNA expression of SGLT1 and PEPT1 in the residual ileum was significantly higher in the blank group than in the normal group (P <0.05). There was no significant difference between the blank group and the GH group and the GLP2 group (P> 0.05). Conclusion Short-term GLP-2 administration may have no significant effect on the mRNA expression of SGLT1 and PEPT1 in the residual ileum of short-term rats.
