观察CD4~+CD25~+Foxp3~+调节性T细胞、IL-10、TGF-β在哮喘和免疫耐受大鼠淋巴液与血液中的水平,探讨其与哮喘及免疫耐受的关系。建立大鼠哮喘和免疫耐受模型,收集激发后0 h、24 h、48 h淋巴液、血液中淋巴细胞,采用流式细胞术检测淋巴液及血液中CD4~+CD25~+Foxp3~+T细胞百分率;采用ELISA方法检测血浆及淋巴液中IL-10、TGF-β水平。结果显示,哮喘组淋巴液和血液的CD4~+CD25~+Foxp3~+T细胞百分率水平、血浆IL-10和TGF-β浓度在各时间点均低于对照组和耐受组(P<0.05);哮喘组和耐受组淋巴液中CD4~+CD25~+Foxp3~+T细胞百分率水平、IL-10浓度在不同时间点均高于血液水平(P<0.05),而TGF-β浓度则低于血液水平(P<0.05);耐受组淋巴液和血液中CD4~+CD25~+Foxp3~+T细胞百分率水平、血浆IL-10和TGF-β浓度与对照组相比无显著性差异(P>0.05)。说明哮喘大鼠淋巴液中存在CD4~+CD25~+Foxp3~+调节性T细胞数量以及功能失调,且淋巴液中CD4~+CD25~+Foxp3~+调节性T细胞水平显著高于其血液水平,而建立免疫耐受后大鼠淋巴液及血液中CD4~+CD25~+Foxp3~+调节性T细胞数量升高,说明通过免疫耐受可以避免哮喘发病或减轻症状,推测通过建立免疫耐受可能成为一种治疗哮喘的有效途径。
To observe the levels of CD4 ~ + CD25 ~ + Foxp3 ~ + regulatory T cells, IL-10 and TGF-β in the lymph and blood of asthmatic and immunocompromised rats, and to explore their relationship with asthma and immune tolerance. The percentage of CD4 ~ + CD25 ~ + Foxp3 ~ + T cells in asthmatic group and tolerated group was higher than that in blood at different time points (P <0.05), while the concentration of TGF-β (P <0.05). The percentages of CD4 ~ + CD25 ~ + Foxp3 ~ + T cells in the lymph and blood of the tolerated group were not significantly different from those in the control group (P> 0.05). The number of CD4 ~ + CD25 ~ + Foxp3 ~ + regulatory T cells and their dysfunction were found in the lymph of asthmatic rats, and the levels of CD4 ~ + CD25 ~ + Foxp3 ~ + regulatory T cells in lymph were significantly higher than those in the blood , While the establishment of immune tolerance rat lymph and blood CD4 ~ + CD25 ~ + Foxp3 ~ + regulatory T cells increased, indicating that by immune tolerance can prevent the incidence of asthma or reduce symptoms, presumably through the establishment of immune tolerance May become an effective way to treat asthma.
