目的:通过研究胃癌患者外周血中FOXP3的表达,初步探讨其在胃癌发病机制中的临床意义。方法:选择胃癌患者与正常对照各31例,用流式细胞仪检测外周血CD4+CD25+T细胞的数量;ELISA检测胃癌患者和正常对照外周血单个核细胞培养上清中Th2型细胞因子白细胞介素(interleukin,IL)-10和Th1型细胞因子IL-2的表达;RT-PCR检测FOXP3的mRNA表达。结果:胃癌患者外周血中CD4+CD25+T细胞约占CD4+T细胞(30.4±1.9)%,显著高于正常人外周血中CD4+CD25+T细胞的比例(3.5±0.8)%(P<0.05);胃癌患者外周血CD4+CD25+T细胞高表达FOXP3;胃癌患者外周血单个核细胞培养上清主要分泌IL-10,而IL-2分泌无显著变化。结论:FOXP3表达的增加对肿瘤免疫具有抑制作用,可能参与胃癌的发生。
Objective: To investigate the clinical significance of FOXP3 in the pathogenesis of gastric cancer by studying the expression of FOXP3 in peripheral blood of patients with gastric cancer. Methods: Thirty-one patients with gastric cancer and normal controls were selected. The number of CD4 + CD25 + T cells in peripheral blood was detected by flow cytometry. The levels of Th2 cytokines in peripheral blood mononuclear cells of gastric cancer patients and normal controls The expression of interleukin (IL) -10 and Th1 cytokine IL-2 were detected by RT-PCR. The mRNA expression of FOXP3 was detected by RT-PCR. Results: The percentage of CD4 + CD25 + T cells in CD4 + T cells (30.4 ± 1.9%) in gastric cancer patients was significantly higher than that in healthy controls (3.5 ± 0.8%) (P <0.05). CD4 + CD25 + T cells were highly expressed in peripheral blood of patients with gastric cancer. FOXP3 was highly expressed in the peripheral blood mononuclear cell culture supernatant of gastric cancer patients, but there was no significant change in IL-2 secretion. Conclusion: The increase of FOXP3 expression can inhibit tumor immunity and may be involved in the occurrence of gastric cancer.
