目的:观察香菇多糖对CMT93细胞的侵袭转移的影响,并对其机制进行初步探讨。方法:CMT93细胞分为对照组和香菇多糖(高、中、低剂量)4组,香菇多糖高、中、低剂量于培养液中加入香菇多糖,使其终浓度分别为0.2g/L、0.4g/L、0.8g/L。分别采用real time PCR、免疫组织化学方法检测肿瘤组织中MMP-2、MMP-9、NF-κB mRNA和MMP-2、MMP-9蛋白含量;采用Boyden小室法观测香菇多糖对CMT93细胞侵袭能力的影响。结果:香菇多糖低剂量能明显降低CMT93细胞中MMP-2、MMP-9、NF-κB mRNA和MMP-2、MMP-9蛋白含量(p<0.05);中、高剂量组能显著降低MMP-2、MMP-9、NF-κB mRNA和MMP-2、MMP-9蛋白含量(p<0.01);低、中、高剂量组能显著降低CMT93细胞的穿膜细胞数(p<0.01)。结论:香菇多糖能降低CMT93细胞的侵袭转移能力,其机制可能与降低NF-κB、MMP-2、MMP-9表达有关。
Objective: To observe the effect of lentinan on the invasion and metastasis of CMT93 cells, and to discuss its mechanism. METHODS: CMT93 cells were divided into control group and lentinan (high, medium and low dose) 4 groups. Lentinan high, medium and low doses were added to broiled lentinan in culture medium so that the final concentrations were 0.2 g/L and 0.4 respectively. g/L, 0.8g/L. Real-time PCR and immunohistochemistry were used to detect the expression of MMP-2, MMP-9, NF-κB mRNA and MMP-2, MMP-9 protein in tumor tissues. The Boyden chamber method was used to observe the invasive ability of lentinan to CMT93 cells. influences. RESULTS: Low dose of lentinan could significantly decrease the protein levels of MMP-2, MMP-9, NF-κB and MMP-2, MMP-9 in CMT93 cells (p<0.05). The medium and high doses could significantly reduce MMP- 2, MMP-9, NF-κB mRNA and MMP-2, MMP-9 protein content (p <0.01); low, medium and high dose group can significantly reduce the number of CMT93 cell membrane cells (p <0.01). Conclusion: Lentinan can reduce the invasion and metastasis of CMT93 cells, and its mechanism may be related to the decrease of NF-κB, MMP-2 and MMP-9 expression.
