参考标准型自动二氧化碳培养箱的结构形式,引入容积法原理计算单位体积内二氧化碳含量,将普通隔水式电热恒温培养箱改装成简易二氧化碳培养箱并进行细胞培养及杂交瘤的建株研究。利用这一装置完成了载脂蛋白B100单克隆抗体杂交瘤细胞系的建株实验,培养的各单细胞株及杂交瘤细胞系均生长良好,平均融合率为66.6%,所获得的单克隆抗体效价高达1:32000。经一年多的实际应用证明,该装置简便经济、性能稳定,克服了传统的干燥缸法
With reference to the standard type of automatic CO2 incubator, the principle of volumetric method was introduced to calculate the carbon dioxide content per unit volume. The ordinary water-isolated electrothermic constant temperature incubator was converted into a simple CO2 incubator and cell culture and construction of hybridomas was studied. Using this device, an apolipoprotein B100 monoclonal antibody hybridoma cell line was established. The cultured single cell lines and hybridoma cell lines grew well with an average fusion rate of 66.6%. The obtained monoclonal antibodies Titer up to 1:32000. After more than one year of practical application, the device is simple, economical, stable, and overcomes the traditional drying cylinder method.
