目的观察清热祛湿益气方对刀豆蛋白A(ConA)诱导的急性肝损伤模型小鼠肝细胞凋亡相关蛋白表达的影响。方法Balb/c雄性小鼠50只,随机分为空白组,模型组,清热祛湿益气方低、中、高剂量组,每组10只,连续灌胃7天。采用Con A诱导复制小鼠急性肝损伤病理模型,观察不同剂量清热祛湿益气方(7.5g/kg、10.0g/kg、12.5g/kg)对肝细胞Bax、Bcl-2以及Caspase-3 mR-NA表达的影响。结果与模型组比较,清热祛湿益气方不同剂量可下调模型小鼠肝组织Bax、Caspase-3 mRNA的表达,并可上调Bcl-2的表达(P<0.05),随剂量增大,Bcl-2阳性率分别为17.56%、18.28%、26.36%。结论清热祛湿益气方可抑制肝细胞凋亡,对Con A诱导的急性肝损伤具有治疗作用。
Objective To observe the effect of Qingre Qushi Yiqi Recipe on hepatocyte apoptosis-related protein expression in ConA-induced acute hepatic injury model mice. Methods Fifty Balb/c male mice were randomly divided into blank group, model group, and low-, medium-, and high-dose groups of Qingre Qushi Qiqi Decoction, 10 in each group. Con A was used to induce the pathological model of acute hepatic injury in mice. The doses of heat-clearing dampness and Qi-invigorating (7.5g/kg, 10.0g/kg, 12.5g/kg) were observed in hepatocyte Bax, Bcl-2, and Caspase-3. Effect of mR-NA expression. Results Compared with the model group, Qingre Qushi Yiqi Formula can reduce the expression of Bax and Caspase-3 mRNA in the liver of model mice, and up-regulate the expression of Bcl-2 (P<0.05). With increasing dose, Bcl The positive rates of -2 were 17.56%, 18.28%, and 26.36%, respectively. Conclusion Qingre Qushi Yiqi prescription can inhibit hepatocyte apoptosis and have therapeutic effect on acute liver injury induced by Con A.
