目的:研究胱硫醚-β-合成酶(CBS)和胱硫醚-γ-裂解酶(CSE)在去势大鼠阴茎海绵体平滑肌(CCSM)中的表达,探讨其在去势后勃起功能障碍发生中的作用。方法:60只8周龄雄性SD大鼠随机分成:假手术2周组(A组)、假手术4周组(B组)、去势2周组(C组)、去势4周组(D组)。术后检测各组血清睾酮(T)水平,免疫组化及RT-PCR检测CBS和CSE在大鼠CCSM中的表达。结果:血清T水平C组[(11.85±6.73)nmol/L]和D组[(1.96±1.23)nmol/L]分别较A组[(89.65±17.13)nmol/L]和B组[(106.75±19.68)nmol/L]显著下降(P<0.05)。CBS、CSE在各组大鼠CCSM中均有表达,CBS mRNA相对表达量C组(0.93±0.14)和D组(0.79±0.17)分别较A组(2.13±0.65)和B组(2.07±0.53)显著降低(P<0.05);CSE mRNA相对表达量C组(0.87±0.20)和D组(0.71±0.12)分别较A组(1.93±0.15)和B组(1.89±0.45)显著降低(P<0.05)。CBS蛋白积分光密度值(IA)C组(130.35±23.56)和D组(80.29±29.65)分别较A组(310.57±130.56)和B组(349.68±112.35)显著降低(P<0.05);CSE蛋白IA C组(93.56±36.64)和D组(58.56±19.95)分别较A组(269.56±116.76)和B组(298.35±100.76)显著降低(P<0.05)。T水平与CBS、CSE在CCSM中的表达水平具有正相关性。结论:T可能通过CBS、CSE的表达调控阴茎勃起功能。
Objective: To investigate the expression of cystathionine-β-synthase (CBS) and cystathionine-γ-lyase (CSE) in ovariectomized rat corpus cavernosum smooth muscle (CCSM) The role of obstacles. Methods: Sixty male Sprague-Dawley rats, aged 8 weeks, were randomly divided into sham operation group (A group), sham operation group (B group), sham operation group (C group) Group D). The levels of serum testosterone (T) in each group were detected after operation. The expressions of CBS and CSE in CCSM were detected by immunohistochemistry and RT-PCR.
