目的:探讨吗啡依赖大鼠脑线粒体细胞色素C氧化酶(COX)活性的变化。方法:采用药参食法培养大鼠吗啡依赖模型,通过差速离心法分离线粒体。采用分光光度及化学发光等方法测定COX活性、心磷脂(CL)含量、膜流动性(MF)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)含量。结果:COX活性、CL含量、MF及SOD活性都明显降低(P<0.01),MDA含量明显升高(P<0.01)。结论:吗啡依赖大鼠脑线粒体COX活性降低,这与SOD活性降低导致的脂质过氧化水平升高有关。
Objective: To investigate the changes of mitochondrial cytochrome C oxidase (COX) activity in brain of morphine dependent rats. Methods: Morphine-dependent model of rats was induced by drug-inducing method and mitochondria were isolated by differential centrifugation. The activity of COX, content of cardiolipin (CL), membrane fluidity (MF), superoxide dismutase (SOD) activity and malondialdehyde (MDA) content were determined by spectrophotometry and chemiluminescence. Results: COX activity, CL content, MF and SOD activity were significantly decreased (P <0.01), MDA content was significantly increased (P <0.01). CONCLUSION: COX activity in mitochondria of morphine-dependent rats is decreased, which is related to the increase of lipid peroxidation induced by the decrease of SOD activity.
