目的:建立母丁香中丁香酚和2-羟基-4,6-二甲氧基-5-甲基苯乙酮的超快速液相色谱(UFLC)含量测定方法,完善母丁香药材的质量标准。方法:采用Shim-pack XR-ODSⅡ(3.0 mm×75 mm,2.2μm),柱温50℃,流动相为甲醇-0.5%磷酸水溶液(60∶40),流速0.8 mL.min-1,检测波长280 nm。结果:丁香酚和2-羟基-4,6-二甲氧基-5-甲基苯乙酮进样量分别在9.98~79.84μg和17.08~136.64μg范围内与峰面积呈良好的线性关系(r=0.9999),平均加样回收率(n=5)分别为99.57%和98.68%。结论:母丁香药材的UFLC含量测定法快速、准确、简便、可靠,是一种值得推广的中药质量评价方法。
OBJECTIVE: To establish a method for the determination of eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone in Camellia oleifera by ultra-fast liquid chromatography (UFLC), and to improve the quality standard of Camellia oleifera. Methods: Shim-pack XR-ODSⅡ (3.0 mm × 75 mm, 2.2 μm) was used. The mobile phase consisted of methanol-0.5% phosphoric acid solution (60:40) at a flow rate of 0.8 mL · min- 280 nm. Results: The injection volume of eugenol and 2-hydroxy-4,6-dimethoxy-5-methylacetophenone showed a good linear relationship with the peak area in the range of 9.98 ~ 79.84μg and 17.08 ~ 136.64μg, respectively r = 0.9999). The average recoveries (n = 5) were 99.57% and 98.68%, respectively. Conclusion: The method for determination of UFLC in Camellia oleifera is fast, accurate, simple and reliable. It is a worthy method to evaluate the quality of traditional Chinese medicine.
