LAK细胞体外诱导过程中要求条件高,易污染,操作烦琐。改进LAK疗法除了提高LAK活性、降低IL-2用量外,缩短LAK体外诱导时间亦是重要方面。本文采用~(125)IUdR释放法测定了大剂量IL-2体外短期诱导人脐血,正常供血者、良性肿瘤、恶性肿瘤病人外周血淋巴细胞的抗肿瘤活性,发现四种来源的淋巴细胞在大剂量IL-2作用15分钟时均可诱导出一定水平的LAK活性。IL-2用量在6×10~3U/1.2×10~7细胞/ml较合适,最适体外培养时间为3天,7天后仍有一定的抗瘤活性、但这种快速LAK活性仍低于单位淋
The in vitro induction process of LAK cells requires high conditions, is easy to pollute, and is cumbersome to operate. In addition to improving LAK activity and reducing the amount of IL-2, improving the LAK in vitro induction time is also an important aspect. In this study, ~125 IUDR release method was used to determine the anti-tumor activity of high-dose IL-2 in peripheral blood, normal blood donors, benign tumors, and malignant tumors. High-dose IL-2 induced a certain level of LAK activity at 15 minutes. The dosage of IL-2 is appropriate at 6×10~3U/1.2×10~7 cells/ml, and the optimal in vitro culture time is 3 days.
