本实验以生理盐水、ｉｌｏｐｒｏｓｔ及其改良液为保存液，在４℃保存兔胎脑皮层组织，以高效液相色谱分析仪（ＨＰＬＣ）测定在保存过程中的腺苷酸含量变化，以了解胎脑组织在４℃保存时的能量代谢情况。结果显示，在生理盐水组及ｉｌｏｐｒｏｓｔ液组，随保存时间延长，胎脑组织中的三磷酸腺苷（ＡＴＡＴＰ）含量、能量负荷（ＥＣ）水平明显下降（Ｐ＜０．０１），在保存２小时接近耗竭，且两组间无明显差异（Ｐ＞０．０５）；而改良ｉｌｏｐｒｏｓｔ液组的ＡＴＰ含量及ＥＣ水平在保存２小时却略高于保存１小时（Ｐ＞０．０５）。此结果表明，要改进４℃保存的效果，不仅需应用细胞保护剂，还应考虑如何延缓能量衰竭的发生。本实验通过在ｉｌｏｐｒｏｓｔ液中加入ＡＴＰ等物质，获得了较ｉｌｏｐｒｏｓｔ液更好的保存效果。
In this experiment, physiological saline, iloprost and its modified solution as the preservation solution, stored at 4 ℃ rabbit cerebral cortex tissue by high performance liquid chromatography (HPLC) determination of adenylate acid content changes in the preservation process to understand the fetal Energy metabolism of brain tissue when stored at 4 ° C. The results showed that in the saline group and the iloprost solution group, the ATATP content and the energy load (EC) level in fetal brain tissue were significantly decreased (P <0.01) with prolongation of storage time, and nearly disappeared after 2 hours of storage (P> 0.05). However, the levels of ATP and EC in the modified iloprost fluid group were slightly higher than those of the control group for 1 hour (P> 0.05). The results show that to improve the effect of 4 ℃ preservation, not only the application of cytoprotective agents, but also consider how to delay the occurrence of energy failure. In this experiment by adding iloprost solution of ATP and other substances, obtained better than iloprost preservation effect.
