目的 :证明上皮生长因子受体 (EGFR)基因与白血病发生和骨髓再生的相关性。方法 :应用鸡原始红细胞增多症病毒癌基因 V - erb B作探针 ,进行 Southern印迹杂交 ,对 8例不同贫血状态下的血液病 ,包括 5例骨髓增生异常综合征 (MDS) ,1例家族性急性髓细胞白血病及其亲属 ,1例缺铁性贫血 (IDA)和 1例范柯尼贫血 (FA)等进行检测 ,并以 7例 AL、MDS和 AA父母为对照。结果 :MDS和 AL有 C- erb B1 重排 /扩增。患者父母仅有 C- erb B1 重排 ,大多数无胚系扩增 ;MDS和 IDA有 C- erb B1 胚系扩增 ,但 IDA无 C- erb B1 重排 ;正常人大多数与其父母一样 ,既无 C- erb B1 胚系扩增 ,也无 C- erb B1 重排。结论 :C- erb B1 即 EGFR基因重排 /扩增与 MDS和 AL有关 ,且其胚系扩增可能还与骨髓再生相关。白血病的发生还可能与骨髓再生有负相关 ,且对 EGF不依赖
PURPOSE: To demonstrate the association of epidermal growth factor receptor (EGFR) gene with leukemia and bone marrow regeneration. Methods: The chicken erythrocytic precursor virus V - erb B gene was used as a probe to carry out Southern blot hybridization. Eight cases of hematological diseases with different anemia including 5 cases of myelodysplastic syndrome (MDS), 1 case of familial Acute myeloid leukemia and its relatives, 1 case of iron deficiency anemia (IDA) and 1 case of Fanconi anemia (FA) were detected in 7 AL, MDS and AA parents. Results: C-erb B1 rearrangement / amplification in MDS and AL. The patients had only C-erb B1 rearrangements, most of them had no germ-line amplification; MDS and IDA had C-erb B1 germline expansion but IDA had no C-erb B1 rearrangement; most of the normal people, like their parents, both No C erb B1 germline expansion nor C erb B1 rearrangement. CONCLUSION: C-erb B1, EGFR gene rearrangement / amplification is associated with MDS and AL, and its germline expansion may be related to bone marrow regeneration.
