目的研究补中益气汤及其配伍对脾气虚大鼠胃泌素(GAS)及GAS mRNA表达的影响,并探讨相关作用机制。方法以大黄、游泳力竭、饥饿三因素复合方法复制脾气虚大鼠模型,造模成功后,将SD大鼠随机分为正常对照组、脾气虚模型组、补中益气汤组、黄芪-升麻、柴胡配伍组和减方组(原方去“黄芪-升麻、柴胡”配伍组),给药14 d后测定大鼠血清GAS含量和GAS mRNA。结果与脾气虚模型组比较,补中益气汤组能有效升高脾气虚大鼠GAS含量(P<0.01)并增强GAS mRNA的表达(P<0.01),而黄芪-升麻、柴胡配伍组和减方组对脾气虚大鼠GAS含量及GAS mRNA的表达影响不大(P>0.05)。结论补中益气汤能显著提高GAS及GAS mRNA的表达来改善脾气虚,推测是其治疗脾气虚证的机制之一,“黄芪-升麻、柴胡”配伍是方中不可或缺的重要组成部分。
Objective To study the effect of Buzhong Yiqi Decoction and its compatibility on gastrin (GAS) and GAS mRNA expression in spleen deficiency rats, and to explore the related mechanism. Methods The rat model of spleen - qi deficiency was established by three - factor combination of rhubarb, exhaustive swimming and starvation. After successful modeling, the SD rats were randomly divided into normal control group, spleen deficiency model group, Buzhongyiqi Decoction group, Astragalus - Cimicifuga, Bupleurum compatibility group and subtraction group (the original prescription to “Astragalus - cohosh, Bupleurum compatibility group), 14 d after administration measured serum GAS content and GAS mRNA. Results Compared with the model group, Buzhongyiqi Decoction could effectively increase the GAS content (P <0.01) and enhance the expression of GAS mRNA (P <0.01) There was no significant difference between GAS content and GAS mRNA expression in rats with spleen deficiency (P> 0.05).
