目的:观察抑癌基因ASPP家族在不同肿瘤细胞株中的表达水平,探讨其在肿瘤的发生、发展及预后中的作用。方法:应用单核细胞分离液分离正常人血液单核细胞,用DMEM培养液培养细胞株jurkat、k-562、HL-60、成纤维细胞、HT-29、所得细胞用Tripure分离试剂提取总RNA。最后用RT-PCR检测ASPPmRNA的表达水平。结果:ASPP1与ASPP2mRNA在正常人细胞中的表达水平比肿瘤细胞株的表达水平高(P<0.05)。iASPP的表达量除在HT-29细胞株中与正常细胞差异无统计学意义(P>0.05),其他肿瘤细胞株的表达量均高于正常细胞的表达量(P<0.01)。结论:抑癌基因ASPP家族的促p53细胞凋亡功能在正常细胞转变为恶性的过程中有重要的作用。
OBJECTIVE: To observe the expression of tumor suppressor gene ASPP family in different tumor cell lines and to explore its role in tumorigenesis, development and prognosis. METHODS: Normal human monocytes were isolated from mononuclear cells by mononuclear cell culture. Cell lines jurkat, k-562, HL-60, fibroblasts and HT-29 were cultured with DMEM. The resulting cells were extracted with Tripure reagent . Finally, the expression of ASPP mRNA was detected by RT-PCR. Results: The expression of ASPP1 and ASPP2 mRNA in normal human cells was higher than that in tumor cell lines (P <0.05). The expression of iASPP was not significantly different from that of normal cells in HT-29 cells (P> 0.05). The expression of iASPP was higher than that in normal cells (P <0.01). Conclusion: ASPP family of tumor suppressor genes promote the apoptosis of p53 cells in normal cells into malignant process has an important role.
