作者用~3H-UR体外掺入后的放射自显影法观察RNA的核-浆转运,观察对象有:10例健康成人骨髓细胞,20例儿童急性原淋巴细胞白血病外周血浓集白细胞,4例原粒细胞白血病患者外周血浓集由细胞,以及经培养三天的9例健康人,8例慢淋白血病人的外周血淋巴细胞。结果:健康人的淋巴细胞与~3H-UR一起孵育1小时后,仅核(主要是核仁)被标记。将细胞洗涤后,转移到无放射活性的培养液中继续培养1小时,胞浆上的标记颗粒即有增加。培养24小时后在全部细胞中都可见到标记物由核转运到胞浆。尤其
The authors observed the nuclear-plasma translocation of RNA using autoradiography after incorporation of ~3H-UR. Observed subjects were: 10 healthy adult bone marrow cells, 20 children with acute promyelocytic leukemia, peripheral blood white blood cells, and 4 patients. Peripheral blood from patients with myeloblastic leukemia was enriched by cells, and 9 healthy individuals who had been cultured for 3 days and peripheral blood lymphocytes from 8 patients with chronic leukemia. Results: After incubation of healthy human lymphocytes with ~3H-UR for 1 hour, only the nucleus (mainly nucleoli) was labeled. After the cells were washed, they were transferred to a non-radioactive culture medium and cultured for 1 hour. The labeled particles on the cytoplasm increased. After 24 hours of culture, the translocation of the label from the nucleus to the cytoplasm was observed in all cells. especially
