目的:建立简便有效、适用于药动学研究的法莫替丁血药浓度高效液相(HPLC)测定方法。方法:采用柱切换HPLC技术。分析柱(150mm×4.6mm,ID)固定相为YWGC18(5μm),分析流动相为甲醇与乙酸铵缓冲液;预处理柱(50mm×4.6mm,ID)填料为YWGC18(10μm),预处理流动相为乙酸铵缓冲液。结果:法莫替丁血浆浓度标准曲线线性范围为5~200ng/ml(r=0.9998),血浆中最低检测质量浓度为4ng/ml。方法日内及日间精密度分别为2.97%±1.56%,6.04%±1.62%,回收率为98.37%±2.91%。结论:应用本方法研究法莫替丁在正常人体内的药动学,具有简便迅速、准确灵敏、易于实现自动化等特点。
Objective: To establish a HPLC method for the determination of famotidine in plasma, which is simple and effective and suitable for pharmacokinetic studies. Methods: Column switching HPLC technique was used. Analysis column (150mm × 4.6mm, ID) The stationary phase was YWGC18 (5μm), and the mobile phase was methanol and ammonium acetate buffer. The pretreatment column (50mm × 4.6mm, ID) was YWGC18 10μm), pretreatment mobile phase ammonium acetate buffer. Results: The standard curve of plasma concentration of famotidine linear range of 5 ~ 200ng / ml (r = 0.9998), the lowest plasma concentration of 4ng / ml. The intra-day and inter-day precision were 2.97% ± 1.56%, 6.04% ± 1.62%, respectively. The recoveries were 98.37% ± 2.91%. Conclusion: This method is used to study the pharmacokinetics of famotidine in normal human body. It is simple, rapid, accurate, sensitive and easy to automate.
