在Mg~(2+)存在下,用(γ-~(32)P)ATP与猪红细胞膜保温(30℃)3min,观察一些药物对猪红细胞膜上磷脂酰肌醇磷酸化的影响。用氯仿:甲醇中止反应。用酸性氯仿:甲醇混合液抽提磷脂及硅胶板TLC分离磷脂。经放射自显影及液体闪烁计数测定生成的(~(32)P)-磷脂酰肌醇-4-磷酸。结果表明①肝素和新霉素对磷脂酰肌醇磷酸化有抑制作用;②六氯环己烷对磷脂酰肌醇磷酸化有促进作用;③Li_2SO_4(10mmol/L)、EGTA(100μmol/L)、茶硷(100μmol/L)作用不明显;④二甲亚矾在低浓度时有促进作用,而在较高浓度时则有抑制作用。结果提示上述作用可能与这些药物的作用机理有关。
The effects of some drugs on the phosphatidylinositol phosphatidylinositol (Phosphatidylinositol Phosphatidylinositol Phosphatidylinositol Phosphatidylinositol Phosphatidylinositol Phosphatidylinositol Phosphatidylinositol Phosphorylation in pig erythrocyte membrane were observed with (γ- (32) P) ATP incubated with porcine erythrocyte membrane (30 ℃) The reaction was quenched with chloroform: methanol. Phospholipids were extracted by TLC using a mixture of acidic chloroform and methanol. (~ (32) P) -Phosphatidylinositol-4-phosphate generated by autoradiography and liquid scintillation counting. The results showed that heparin and neomycin inhibited the phosphoinositide phosphorylation; ② HCH promoted the phosphatidylinositol phosphorylation; ③Li_2SO_4 (10mmol / L), EGTA (100μmol / L) Theophylline (100μmol / L) had no obvious effect. (4) Dimethylsulfoxide promoted the proliferation at low concentration and inhibited at higher concentration. The results suggest that these effects may be related to the mechanism of action of these drugs.
