目的 探讨大肠癌、大肠腺瘤及正常粘膜中nm 2 3 H 1,p16蛋白的表达。方法 应用免疫组化ABC法对 3 0例大肠癌、2 5例大肠腺瘤及 2 5例正常大肠粘膜中nm 2 3 H 1、p16蛋白的表达进行测定。 结果 大肠癌组织中nm 2 3 H 1与 p16阳性表达率分别为 66.7%和 40 .0 % ;p16在大肠癌中的表达水平明显低于正常大肠粘膜及腺瘤中的表达 (P <0 .0 5 ) ,而nm 2 3 H 1表达在各组间虽呈下降趋势 ,但无显著性差异 (P >0 .0 5 ) ;两者在大肠癌中的表达存在相关关系 (P <0 .0 5 ) ,并与癌组织的分化程度、有无淋巴结转移及患者的生存时间等均有明显相关性 (P <0 .0 5 )。结论 抑癌基因 p16的失活、nm 2 3 H 1基因的低表达可能与大肠癌的发生、发展、转移及预后有关。
Objective To investigate the expression of nm23H1 and p16 protein in colorectal carcinoma, colorectal adenoma and normal mucosa. Methods Immunohistochemical ABC method was used to detect the expression of nm23H1 and p16 protein in 30 colorectal carcinomas, 25 colorectal adenomas and 25 normal colorectal mucosa. Results The positive rates of nm23 H 1 and p16 in colorectal carcinoma were 66.7% and 40.0%, respectively. The expression of p16 in colorectal carcinoma was significantly lower than that in normal colorectal mucosa and adenoma (P <0.05). The expression of nm 2 3 H 1 in all the groups showed a decreasing trend, but no significant difference (P> 0.05). There was a correlation between the expression of nm 2 3 H 1 and colorectal carcinoma (P 0. 05). There was a significant correlation between tumor differentiation and lymph node metastasis and survival time (P <0.05). Conclusion The inactivation of tumor suppressor gene p16 and the low expression of nm 2 3 H 1 gene may be related to the occurrence, development, metastasis and prognosis of colorectal cancer.
