苯妥英是治疗癫痫的首选药物,缺点是口服后吸收不完全且不规则,个体的生物半衰期差异大。本文选择尿素、聚乙二醇(PEG)6000和聚乙烯吡咯烷酮(PVP)为载体,分别与苯妥英制成固态分散体,研究该分散体的溶解度和溶出度。样品为10%苯妥英和90%载体的固态分散体和物理混合物。其中PEG和尿素的固态分散体用熔融法制备,PVP用溶剂法制备。将上述分散体或物理混合物制成三种片剂,处方A含原料药,以氯化钠为载体;处方B为药物与载体的物理混合物;处方C为药
Phenytoin is the drug of choice for the treatment of epilepsy, the disadvantage is incomplete and irregular absorption after oral administration, the individual biological half-life difference. In this paper, urea, polyethylene glycol (PEG) 6000 and polyvinylpyrrolidone (PVP) were chosen as carriers to prepare solid dispersions with phenytoin respectively. The solubility and dissolution of the dispersions were studied. The sample is a solid dispersion of 10% phenytoin and 90% carrier and a physical mixture. The solid dispersions of PEG and urea were prepared by melt method and PVP by solvent method. The above dispersion or physical mixture is made into three kinds of tablets. The prescription A contains the raw material medicine and sodium chloride is used as the carrier; the prescription B is the physical mixture of the medicine and the carrier; the prescription C is the medicine
