目的:研究天麻不同种质DNA指纹图谱,探讨扩增酶切片段长度多态性(AFLP)、简单序列重复(SSR)分子遗传标记技术在天麻遗传变异分析上的应用。方法:采用AFLP、SSR及其聚类树对24份不同天麻种质材料的遗传多样性等进行分析,并对这2种分子标记进行比较。结果:检测到AFLP多态性44%-89%、SSR多态性15%-69%;AFLP聚类相似性系数0.56-0.98,SSR聚类相似性系数0.13-1.00。2种分子标记将天麻变型种质聚类划分的结果相近,但SSR能将野生天麻聚在一类。结论:天麻不同种质AFLP多态性高于天麻SSR,SSR对野生天麻有较强的区分能力。
Objective: To study DNA fingerprinting of different germplasms of Gastrodia elata Blume, and to explore the application of AFLP and SSR molecular genetic markers in genetic variation analysis of Gastrodia elata Blume. Methods: The genetic diversity of 24 different Gastrodia elata varieties was analyzed by AFLP, SSR and cluster tree. The two molecular markers were compared. Results: AFLP polymorphism was detected in 44% -89%, SSR polymorphism was 15% -69%, AFLP clustering similarity coefficient was 0.56-0.98, and SSR clustering similarity coefficient was 0.13-1.00.2. Variation of germplasm clustering results are similar, but SSR can gather wild Gastrodia elata species. CONCLUSION: AFLP polymorphism of different genotypes of Gastrodia elata Blume is higher than that of Gastrodia elata Blume, SSR has a strong ability to distinguish between them.
