星形胶质细胞之间通过缝隙连接形成偶联,其缝隙连接蛋白为Cx43。该缝隙连接对神经元兴奋时释放到细胞外的K+起空间缓冲作用。本研究以遗传性癫痫易感大鼠P77PMC为模型,采用原位杂交的方法,观察了一次听源性惊厥后脑内星形胶质细胞Cx43基因表达的变化,结果显示,P77PMC大鼠一次听源性惊厥后在大脑皮质、海马Cx43mRNA表达呈时间依赖性增加,从惊厥后2h开始增加,4~8h达高峰,24h仍高于对照。结果表明:惊厥时,皮质和海马星形胶质细胞Cx43基因表达增强,可能有利于星形胶质细胞对神经元兴奋时释放到细胞外的K+产生空间缓冲作用,以便维持神经辕围的的K+平衡。
Astrocytes are connected by a gap junction, the connexin is Cx43. This gap junction provides spatial cushion of K + released to the extracellular cells upon neuronal excitation. In this study, epilepsy-susceptible rat P77PMC was used as a model to observe the changes of Cx43 gene expression in astrocytes after auditory convulsions using in situ hybridization. The results showed that the primary source of P77PMC The expression of Cx43mRNA in cerebral cortex and hippocampus increased in a time-dependent manner after sexual convulsion. It increased from 2h after convulsion and reached the peak at 4 ~ 8h, which was still higher than the control at 24h. The results showed that the enhanced expression of Cx43 gene in cortex and hippocampus astrocytes during convulsions may be beneficial to the space buffer effect of astrocytes on releasing K + extracellularly when excited by neurons in order to maintain the nerve K + balance.
