目的:检测通用转录因子3(BTF3)在卵巢癌生长增殖中的作用,探讨其作为治疗靶点的可行性。方法:构建shBTF3慢病毒载体后转染人卵巢癌SKOV3细胞,采用细胞成像仪检测细胞生长状况,流式细胞术检测细胞周期及凋亡率,MTT法检测慢病毒转染后SKOV3细胞增殖状态的改变。结果:shBTF3慢病毒载体成功转染SKOV3细胞后,与对照组相比,实验组细胞生长状况下降,增殖减慢。SKOV3细胞周期G_1期被抑制,细胞凋亡率升高(P<0.05)。结论:BTF3对卵巢癌细胞SKOV3的生长增殖具有促进作用,考虑BTF3基因在卵巢癌中的表达可能与其发生发展及分级分期密切相关。
Objective: To investigate the role of universal transcription factor 3 (BTF3) in the proliferation and proliferation of ovarian cancer and to explore its feasibility as a therapeutic target. Methods: Human ovarian cancer SKOV3 cells were transfected with lentiviral vector shBTF3, the cell growth was detected by cell imager, the cell cycle and apoptosis rate were detected by flow cytometry, and the proliferation status of SKOV3 cells was detected by MTT assay change. Results: After transfection of SKOV3 cells with shBTF3 lentiviral vector, the cell growth and the proliferation of the experimental group decreased compared with the control group. The G1 phase of SKOV3 cell cycle was inhibited and the apoptosis rate was increased (P <0.05). Conclusion: BTF3 can promote the growth and proliferation of ovarian cancer cell line SKOV3. Considering the expression of BTF3 gene in ovarian cancer, it may be closely related to its development and grading and staging.
