目的:观察肿节风复方含药血清对人肝癌细胞HepG2细胞增殖及凋亡的影响。方法:实验设阴性对照组(NS)、5-氟尿嘧啶(5-FU)组(0.025 g·kg-1)和肿节风复方低、中、高剂量组(2.5,5,10 g·kg-1),灌服于大鼠后,提取含药血清。取上述含药血清分别作用于HepG2细胞24,48,72 h;MTT法测定细胞增殖抑制率;ELISA法测定端粒酶活性;荧光显微法及流式细胞术(Annexin V-FITC/PI法)检测肿节风复方含药血清对HepG2细胞凋亡的影响。结果:与对照组比较,肿节风复方含药血清低、中、高剂量组均能抑制HepG2细胞的增殖(高剂量组抑制率最高达42.5%),呈现与时间-剂量依赖趋势;肿节风中、高剂量组能有效抑制HepG2细胞端粒酶活性,与对照组比较P<0.05;肿节风复方含药血清具有诱导HepG2细胞凋亡的作用,与对照组比较P<0.05,其中中剂量组作用24,48,72 h细胞凋亡率分别为(14.2±0.9)%,(30.6±1.0)%,(61.6±1.5)%。结论:肿节风复方含药血清能抑制HepG2细胞增殖、端粒酶活性,诱导HepG2细胞凋亡。
OBJECTIVE: To observe the effect of the drug-containing serum of ZJT on the proliferation and apoptosis of human hepatoma HepG2 cells. Methods: The experimental groups were divided into 5 groups: negative control group (NS), 5-fluorouracil group (0.025 g · kg -1) 1), drench in rats, extract containing serum. The drug-containing serum was applied to HepG2 cells for 24, 48, and 72 h respectively. The inhibition rate of cell proliferation was determined by MTT assay. The telomerase activity was determined by ELISA. The expression of telomerase activity was detected by fluorescence microscopy and flow cytometry (Annexin V-FITC / PI) ) To detect the effect of ZJJP on the apoptosis of HepG2 cells. Results: Compared with the control group, the tumor-containing compound of ZJJH could inhibit the proliferation of HepG2 cells (the highest inhibition rate was 42.5%) in a dose-dependent and time-dependent manner The wind and high dose groups could effectively inhibit the telomerase activity of HepG2 cells, compared with the control group (P <0.05). The drug-containing serum from the JieFengFeng group had the effect of inducing HepG2 cell apoptosis, compared with the control group (P <0.05) The apoptotic rate of 24,48,72 h in dose group were (14.2 ± 0.9)%, (30.6 ± 1.0)% and (61.6 ± 1.5)%, respectively. Conclusion: The drug-containing serum of ZJJH can inhibit HepG2 cell proliferation and telomerase activity and induce HepG2 cell apoptosis.
