目的:建立紫草闪式提取醇提物的HPLC指纹图谱,全面完整地反映其内在化学信息,为紫草醇提物的质量控制提供依据。方法:采用高效液相色谱法,色谱柱为Kromasil 100-5 C18(4.6 mm×250 mm,5μm),以水-甲酸(300∶0.5)-乙腈为流动相进行梯度洗脱,检测波长275 nm,流速1 mL·min-1,柱温25℃。结果:建立了10批紫草闪式提取醇提物的HPLC指纹色谱图,获得了13个共有峰,并指认了其中的3个色谱峰,4,6,10号分别被指认为左旋紫草素、乙酰紫草素和β,β’-二甲基丙烯酰阿卡宁。结论:该方法简便,精密度、重复性和稳定性良好,能有效控制紫草醇提物的质量,为进一步药效研究、制剂研发提供质量保证。
OBJECTIVE: To establish a HPLC fingerprint of the extract of Lithospermum officinalis L., to fully and completely reflect its internal chemical information and provide the basis for the quality control of Lithospermum alcohol extract. Methods: The elution was performed on a Kromasil 100-5 C18 column (4.6 mm × 250 mm, 5 μm) using gradient elution with water-formic acid (300:0.5) -acetonitrile as the mobile phase. The detection wavelength was 275 nm , Flow rate 1 mL · min-1, column temperature 25 ℃. Results: The HPLC fingerprints of 10 batches of Lithospermum officinalis L. extract were established. 13 common peaks were identified and 3 of them were identified, while the 4, 6 and 10 were identified as Rhizoma Coptidis Acetobrotin and beta, beta’-dimethylacryloracan. Conclusion: The method is simple, accurate, reproducible and stable, and can effectively control the quality of the alcohol extract of Lithospermum, which will provide quality assurance for further research on drug efficacy and formulation research and development.
