目的:探讨衰老雌性大鼠性激素及其受体的变化情况和何首乌饮延缓卵巢组织衰老的作用机制。方法:D-半乳糖连续腹腔注射致亚急性衰老动物模型,造模后灌胃何首乌饮连续60天后,大鼠断头取垂体、卵巢。采用全自动微粒子化学发光免疫分析方法检测各组大鼠血清LH,E2,FSH的含量;原位杂交方法检测各组大鼠卵巢细胞FSHR和LHR的变化;West-ern Bloting法检测各组大鼠卵巢ER的变化。结果:同正常对照组相比,模型组大鼠垂体指数和卵巢指数均明显降低,血清E2含量明显降低,LH、FSH升高,ER、FSHR、LHR的表达明显降低,差异有统计学意义(P<0.05)。与模型组比较,何首乌饮组垂体指数和卵巢指数均明显增高,血清E2含量明显升高,LH、FSH降低,ER、FSHR、LHR的表达明显升高,呈现一定的剂量依赖性。结论:衰老时大鼠卵巢正常功能被破坏,何首乌饮通过对性激素和其受体的调节而延缓卵巢的衰老。
Objective: To investigate the changes of sex hormones and their receptors in aging female rats and the mechanism of action of Polygonum multiflorum Thunb on inhibiting ovarian tissue senescence. Methods: The animal model of subacute aging was induced by continuous intraperitoneal injection of D-galactose. After 60 days of continuous gavage with Polygala tenuifolia, the pituitary and ovary were decapitated in rats. The levels of LH, E2 and FSH in serum of rats in each group were detected by automatic particle chemiluminescence immunoassay. The changes of FSHR and LHR in ovaries were detected by in situ hybridization. Changes in ovarian ER. Results: Compared with the normal control group, the pituitary index and ovarian index in the model group were significantly decreased, the serum E2 level was significantly decreased, LH and FSH levels were increased, the expression of ER, FSHR and LHR was significantly decreased, the difference was statistically significant P <0.05). Compared with the model group, the pituitary index and the ovarian index of the Polygonum multiflorum group were significantly increased, the serum E2 level was significantly increased, LH, FSH decreased, the expression of ER, FSHR, LHR increased significantly, showing a dose-dependent manner. CONCLUSION: The normal function of ovary in rats is destroyed during aging. Polygonum multiflorum drink delays the ovarian aging through the regulation of sex hormone and its receptor.
