目的:比较新型牙髓治疗材料i Root BP Plus与矿物三氧化物凝聚体(mineral trioxide aggregate,MTA)对人牙龈成纤维细胞的细胞毒性。方法:采用四甲基偶氮唑盐(methyl-thiazol-tetrazolium,MTT)法测定上述2种材料不同浸提时间(1、3、7 d)的浸提液原液及不同浓度稀(1:2、1:5)释液对体外培养的人牙龈成纤维细胞增殖的影响。细胞增殖率以x±s表示,采用SPSS20.0软件包对数据进行析因设计的方差分析。结果:i Root BP Plus和MTA的浸提液原液及不同浓度稀释液作用后的细胞增殖率界于77.31%~113.82%,细胞毒性分级为0或1级,评价结果为无细胞毒性。不同时间点、不同稀释度下,2种材料对体外培养的人牙龈成纤维细胞增殖率的影响无显著差异(F浓度*时间*材料=1.393,P=0.256)。结论 :i Root BP Plus与MTA均对体外培养的人牙龈成纤维细胞无细胞毒性。
OBJECTIVE: To compare the cytotoxicity of i Root BP Plus and mineral trioxide aggregate (MTA) on human gingival fibroblasts. Methods: The extracts of the extracts of the two materials at different extraction times (1, 3, 7 d) and the different concentrations of dilute solutions (1: 2) were determined by methyl thiazol-tetrazolium , 1: 5) on the proliferation of cultured human gingival fibroblasts in vitro. Cell proliferation rate to x ± s that the use of SPSS20.0 package data analysis of factorial analysis of variance. Results: The cell proliferation rate of i Root BP Plus and MTA extracts was 77.31% ~ 113.82% after treated with different concentrations of stock solutions and cytotoxicity grade 0 or 1. The results showed no cytotoxicity. At different time points and different dilutions, there was no significant difference in the proliferation rate of human gingival fibroblasts cultured in vitro (F concentration * time * material = 1.393, P = 0.256). Conclusion: Both i Root BP Plus and MTA are cytotoxic to human gingival fibroblasts cultured in vitro.
