目的 :通过观察人甲状旁腺素 (hPTH1 3 4 )及其聚乙二醇修饰物对鸡血钙含量的影响判断它们的体内活性。方法 :12日龄的小鸡分别翼静脉注射赋形剂 ,hPTH1 3 4 ,Cys(mPEG50 0 0 MAL) hPTH1 3 4 和hPTH1 3 4 Cys(mPEG50 0 0 MAL) NH2 。 6 0min和 12 0min后翼静脉采血 ,分离血清并测定钙含量。结果 :给药后 6 0min ,hPTH1 3 4 ,Cys(mPEG50 0 0 MAL) hPTH1 3 4 和hPTH1 3 4 Cys(mPEG50 0 0 MAL) NH2 组静脉血清钙含量均明显升高 ;12 0min后 ,Cys(mPEG50 0 0 MAL) hPTH1 3 4 组血清钙含量仍维持升高水平 ,而hPTH1 3 4 和hPTH1 3 4 Cys(mPEG50 0 0 MAL) NH2 组血清钙含量则明显回落。结论 :合成人甲状旁腺素 (hPTH1 3 4 )及其聚乙二醇修饰物可以明显升高小鸡的血清钙水平 ;N端聚乙二醇修饰的hPTH1 3 4 维持血钙升高的时间较长。
OBJECTIVE: To determine their in vivo activity by observing the effect of human parathyroid hormone (hPTH1 3 4) and its polyethylene glycol modification on chicken blood calcium levels. Methods: 12-day-old chickens were injected with vehicle, hPTH1 34, Cys (mPEG50 0 MAL) hPTH1 34, and hPTH1 34 Cys (mPEG50 0 MAL) NH2 respectively. Blood was collected from the wing vein at 60 min and 120 min, the serum was separated and the calcium content was determined. Results: After 60 min of administration, the serum calcium levels of hPTH1 3 4, Cys (mPEG50 0 0 MAL) hPTH1 3 4 and hPTH1 3 4 Cys (mPEG50 0 0 MAL) NH2 groups were significantly increased. After 120 min, Cys ( Serum calcium levels in hPTH1 34 group were still elevated, but serum calcium levels in hPTH1 34 and hPTH1 34 Cys (mPEG50 0 0 MAL) NH2 group were significantly decreased. CONCLUSION: The synthetic human parathyroid hormone (hPTH1 3 4) and its polyethylene glycol modified substance can significantly increase the serum calcium level in chickens. The N-terminal polyethylene glycol modified hPTH1 3 4 maintains the elevated serum calcium Longer.
