目的研究中药活性成分柠檬醛对实验小鼠侵袭性肺曲霉病(IPA)的作用。方法小鼠随机分成正常组,正常+接种烟曲霉菌组,IPA模型组,IPA+不同剂量(高、中、低3个剂量)柠檬醛给药组及阳性对照组。建立IPA模型后,ip给药不同剂量柠檬醛,阳性对照组ig酮康唑,连续7 d,第8天处死各组小鼠,分别取肺、脾、肝、肾组织行烟曲霉菌培养和观察病理损伤;同时另取各用药组和IPA模型组15只小鼠做生存实验。结果中、高剂量柠檬醛治疗的IPA小鼠各脏器烟曲霉菌落数均低于IPA模型小鼠,且烟灰色色素明显减少;病理结果显示IPA模型小鼠肺组织可见大量菌丝,炎症细胞浸润,病理损伤明显;中、高剂量柠檬醛组肺组织未见菌丝。生存试验表明柠檬醛中、高剂量组小鼠的生存率和生存质量明显优于未用药组。结论柠檬醛可缓解IPA小鼠的肺组织病理损伤,明显减缓IPA的发展。
Objective To study the effect of citral, an active component of traditional Chinese medicine, on invasive pulmonary aspergillosis (IPA) in mice. Methods Mice were randomly divided into normal group, normal + inoculation group of Aspergillus fumigatus group, IPA model group, IPA + different doses (high, medium and low doses) of citral administration group and positive control group. After the IPA model was established, different doses of citral were administered ip and ce ketoconazole was administered as a positive control group. After 7 consecutive days, mice in each group were sacrificed on the 8th day, and lung, spleen, liver, and kidney tissues were cultured for Aspergillus fumigatus respectively. Observe the pathological lesions; at the same time, another 15 mice from each drug group and IPA model group were used for survival experiments. Results The colony counts of Aspergillus fumigatus in IPA mice treated with medium- and high-dose citral were lower than those in IPA mice, and the grey-pigment pigments were significantly reduced. The pathological results showed that there were a lot of hyphae and inflammatory cells in the lungs of IPA model mice. Infiltration, pathological damage was obvious; no mycelium was observed in the lung tissues of the middle and high doses of citral. The survival test showed that the survival rate and quality of life of the middle and high doses of citral mice were significantly better than those of the untreated group. Conclusion Citral can alleviate the pathological injury of lung tissue in IPA mice and significantly slow down the development of IPA.
