目的分析血水草生物碱(ECA)致钉螺肝脏蛋白质表达的变化。方法10mg/LECA浸泡钉螺36h后,解剖活钉螺并收集肝脏,双向电泳分离ECA组和清水对照组钉螺肝脏蛋白质,ImageMaster2D5.0软件分析图谱,筛选差异蛋白质,并用MALDI-TOF-TOF质谱进行鉴定。结果ECA组和清水对照组分别检出(354±110)、(311±12)个蛋白点;质谱鉴定获得假定蛋白、ENSANGP00000022175蛋白、醛脱氢酶、未知蛋白、ENSAN-GP00000027067蛋白、类似锚蛋白3的1亚型4亚亚型蛋白、相似A型肝炎病毒短型细胞受体1的蛋白、线粒体异柠檬酸脱氢酶2样蛋白、类似角蛋白的蛋白、角蛋白10、角蛋白1共11个蛋白质,这些蛋白质均为ECA处理后表达上调的蛋白质。结论ECA可引起钉螺肝脏蛋白质表达发生改变。
OBJECTIVE: To analyze the changes of protein expression in the liver of Oncomelania hupepensis induced by alkaloids from blood plants. Methods After the snails were soaked in 10 mg / L ECA for 36 h, the liver was dissected and the liver was collected. Two-dimensional electrophoresis was used to separate the liver proteins from the ECA group and the water control group. ImageMaster2D5.0 software was used to analyze the differential proteins and identified by MALDI-TOF-TOF mass spectrometry. The hypothetical protein, ENSANGP00000022175 protein, aldehyde dehydrogenase, unknown protein, ENSAN-GP00000027067 protein, 3 subtype 1 subtype 4 subtype, a protein similar to hepatitis A virus short form cell receptor 1, mitochondrial isocitrate dehydrogenase 2-like protein, keratin-like protein, keratin 10, keratin 1 Eleven proteins, all of these proteins are upregulated after ECA treatment. Conclusions ECA can cause hepatic protein expression in the snail to change.
