采用蔗糖等密度梯度离心法,从T-S-Z系统的SRSV/3T3,L783V/3T3,L615K和A9细胞系的培养上清液及L6565小鼠淋巴细胞白血病组织的无细胞提取液中分离小鼠白血病病毒(MuLV)。应用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)方法,将SRS-MuLV,L783-MuLV,L615K-MuLV,L6565-MuLV和Mo-MuLV进行蛋白组成比较研究。结果表明,SRS-MuLV,L783-MuLV,L615K-MuLV,L6565-MuLV和Mo-MuLV的p30电泳迁移率不尽相同。T-S-Z系统的MuLV均有明显的gp45存在,与Mo-MuLV间存在差异。上述结果说明我国的T-S-Z系统中的MuLV是具有自身特点的,但彼此之间又存在细小差异,与国外的Mo-MuLV相比是亲缘关系不同的小鼠白血病病毒株。
Mouse leukemia virus was isolated from cell culture supernatants of SRSV / 3T3, L783V / 3T3, L615K and A9 cell lines of TSZ system and cell-free extracts of L6565 mouse lymphocytic leukemia tissue using density gradient centrifugation such as sucrose MuLV). The protein compositions of SRS-MuLV, L783-MuLV, L615K-MuLV, L6565-MuLV and Mo-MuLV were compared using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) The results showed that the p30 electrophoretic mobility of SRS-MuLV, L783-MuLV, L615K-MuLV, L6565-MuLV and Mo-MuLV varied notably. The MuLV of T-S-Z system had obvious gp45, which was different from that of Mo-MuLV. The above results show that MuLV in our T-S-Z system has its own characteristics, but there are minor differences between them. Compared with Mo-MuLV in foreign countries, MuLV is a mouse leukemia virus strain with different genetic relationship.
