本文提出了一种用于研究伴刀豆球蛋白 A 及其与某些多糖结合反应的简单、迅速而特异的单扫极谱方法。在含有0.001mol/L 2,2′一联吡啶、0.015mol/L 6次甲基四胺及0.1mol/L 氯化钠溶液中,泮刀豆球蛋白 A 显示一单扫极谱波,阴极峰电位是-1.50V(对饱和甘汞电极)。在1.0×10~(-8)mol/L~1.2×10~(-7)mol/L 浓度范围内,用导数单扫极谱观察到峰电流随伴刀豆球蛋白 A 的浓度呈线性变化。对伴刀豆球蛋白 A 的极谱波的性质和机理作了初步探讨。此外,本文表明单扫极谱可以成为在溶液中研究伴刀豆球蛋白 A 与其互补多糖结合反应的有效手段。
In this paper, a simple, rapid and specific single-sweep method for the determination of concanavalin A and its interaction with some polysaccharides has been proposed. In concanavalin A containing 0.001mol / L 2,2’-bipyridyl, 0.015mol / L 6-methylamine and 0.1mol / L sodium chloride solution, The peak potential is -1.50V (for saturated calomel electrode). In the concentration range of 1.0 × 10 -8 mol / L to 1.2 × 10 -7 mol / L, the peak current was linearly changed with concanavalin A by using derivative single sweep polarography. The properties and mechanism of polarographic wave of concanavalin A were preliminarily discussed. In addition, this paper shows that single-sweep polarography can be an effective method to study concanavalin A binding to its complementary polysaccharide in solution.
