目的提取紫菜中类菌孢素氨基酸(Mycosporine-like amino acids,MAAs),探究MAAs对外源性自由基的清除及对内源性抗氧化酶的活力影响。方法采用乙醇提取、层析纯化的方法对紫菜MAAs进行提取纯化;利用液质联用法鉴定紫菜MAAs的种类及结构;采用邻苯三酚-鲁米诺、Fenton反应法和DPPH法研究紫菜MAAs对自由基的清除作用,并测定总抗氧化能力;选取正常ICR小鼠,随机分为空白对照组(NC)、阳性对照组(MP)以及MAAs给药组(MAAs),通过对比各组小鼠血清中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活力及丙二醛(MDA)的含量,对MAAs化合物体内抗氧化活性进行研究。结果紫菜中2种主要的MAAs成分为:Porphyra-334和shinorine;MAAs化合物及Porphyra-334组分具有显著的自由基清除作用,其中功能性成分为Porphyra-334。MAAs的总抗氧化能力随浓度的增大呈现增强趋势;与对照组相比,给药组小鼠血清中SOD、GSH-Px的活力显著提升(P<0.05)。结论紫菜MAAs具有显著的抗氧化活性。
Objective To extract Mycosporine-like amino acids (MAAs) from seaweed to explore the effect of MAAs on the removal of exogenous free radicals and the activity of endogenous antioxidant enzymes. Methods The MAAs from seaweed were extracted and purified by ethanol extraction and chromatographic purification. The species and structure of MAAs were identified by LC-MS method. The effects of MAAs on seaweed were studied by pyrogallol-luminol, Fenton reaction and DPPH method. Free radical scavenging activity and determine the total antioxidant capacity. Normal ICR mice were randomly divided into blank control group (NC), positive control group (MP) and MAAs administration group (MAAs) The activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and the content of malondialdehyde (MDA) in serum were determined to study the antioxidant activity of MAAs in vivo. Results The two major MAAs in seaweed were Porphyra-334 and shinorine. MAA and Porphyra-334 had significant free radical scavenging activity. The functional components were Porphyra-334. The total antioxidant capacity of MAAs tended to increase with increasing concentration. Compared with the control group, the activities of SOD and GSH-Px in the serum of the treated mice significantly increased (P <0.05). Conclusion seaweed MAAs has significant anti-oxidative activity.
