培养的牛心内膜内皮细胞 (bovineendocardialendothelialcells ,BECEC) ,用 2mmol/ml硝普钠 (sodi umnitroprusside,SNP)损伤 2 4h ,出现典型的病理性凋亡。在 0 2mg/ml山莨菪碱加入培养基中 30min后 ,再加入SNP( 2mmol/ml) ,共同孵育 2 4h。发现山莨菪碱保护组与SNP损伤组比较 ,凋亡细胞减少 ;琼脂糖电泳未出现明显的“梯状”条带 ;DNA断裂百分率也降低。结果证明 :山莨菪碱能减轻SNP所致牛心内膜内皮细胞凋亡。
The cultured bovine endocardial endothelial cells (BECEC) were treated with 2 mmol / ml sodiumnitroprusside (SNP) for 24 h. Typical pathological apoptosis occurred. After 0 2 mg / ml anisodamine was added to the medium for 30 min, SNP (2 mmol / ml) was added and incubated for 24 h. Found that anisodamine protective group and SNP injury group, apoptotic cells decreased; agarose electrophoresis showed no obvious “ladder” bands; DNA breakage percentage also decreased. The results show that: Anisodamine can reduce SNP-induced apoptosis of endothelial cells in bovine endocardium.
