平滑肌对维持机体的呼吸、血液循环、消化、生殖等生理功能有重要作用。离体平滑肌制备的反应单一,药物在作用部位的浓度和溶液里的浓度相关性好,是生理、药理科研及教学工作常用的模型。不同种属或不同器官的平滑肌特性差异很大,但作为一种实验手段,理想的平滑肌制备应具有以下特征;(1)含有丰富的平滑肌细胞 这些细胞平行排列形成一薄层,以便于药物或离子的扩散。(2)在同一动物可取得二条同样的肌肉,分别用于对照和测试组,以减少个体差异的影响。(3)若为研究植物神经对肌肉的作用,该制备应具有丰富的植物神经支配。
