目的:初步探讨EGCG对卵巢癌HO-8910细胞增殖的抑制作用及其机制。方法:通过绘制细胞生长曲线、平皿克隆和软琼脂集落形成实验观察EGCG对HO-8910细胞增殖的抑制作用;Western-blotting检测AKT1、Mdm-2与p53蛋白的表达。结果:(1)细胞生长曲线、平皿克隆和软琼脂集落形成实验结果显示,EGCG可有效抑制HO-8910细胞的增殖(n=3,P<0.05)。(2)Westernblotting检测结果显示,EGCG处理后AKT1与Mdm-2蛋白表达均降低,而p53蛋白表达升高(P<0.05)。结论:EGCG通过抑制HO-8910细胞中AKT1与Mdm-2蛋白表达,促使p53蛋白表达而发挥其对细胞增殖的抑制作用。
Objective: To investigate the inhibitory effect of EGCG on the proliferation of ovarian cancer HO-8910 cells and its mechanism. Methods: The inhibitory effects of EGCG on the proliferation of HO-8910 cells were observed by cell growth curve, plate clone and soft agar colony formation assay. The expressions of AKT1, Mdm-2 and p53 proteins were detected by Western-blotting. Results: (1) Cell growth curve, plate clone and soft agar colony formation assay showed that EGCG could effectively inhibit the proliferation of HO-8910 cells (n = 3, P <0.05). (2) The results of Western blotting showed that the protein expressions of AKT1 and Mdm-2 decreased and the expression of p53 protein increased after EGCG treatment (P <0.05). CONCLUSION: EGCG exerts its inhibitory effect on cell proliferation by inhibiting the expression of AKT1 and Mdm-2 in HO-8910 cells and promoting the expression of p53 protein.
