目的:建立HPLC法测定苦豆子中野靛碱和槐胺碱的含量。方法:采用Waters x-Bridge C18(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-含2.0 mL.L-1三乙胺的0.05 mol.L-1的磷酸二氢钾溶液(90∶10),流速为1.0 mL.min-1,检测波长为311 nm。结果:野靛碱和槐胺碱的线性范围分别为4.972~165.7μg(r=0.9994)和4.388~146.3μg(r=0.9991);平均回收率(n=9)分别为101.8%和99.3%。结论:本方法简便、可靠、准确,可为苦豆子药材的开发利用提供理论依据。
Objective: To establish an HPLC method for the determination of podophylline and sophoramine in Sophora alopecuroides. Methods: A Waters x-Bridge C18 (4.6 mm × 250 mm, 5 μm) column was used. The mobile phase consisted of acetonitrile-containing 0.05 mol·L -1 potassium dihydrogen phosphate solution containing 2.0 mL.L-1 triethylamine : 10), the flow rate was 1.0 mL.min-1, the detection wavelength was 311 nm. Results: The linear ranges of indomethacin and sophoramine were 4.972-165.7 μg (r = 0.9994) and 4.388-146.3 μg (r = 0.9991), respectively. The average recoveries (n = 9) were 101.8% and 99.3%, respectively. Conclusion: The method is simple, reliable and accurate, which can provide theoretical basis for the exploitation and utilization of Sophora alopecuroides.
