目的探讨硫辛酸(LA)对帕金森大鼠黑质多巴胺能神经细胞存活及炎症介质的影响。方法选用健康老年雄性SD30只,随机分为对照组、损伤组和治疗组;采用立体定向技术向损伤组和治疗组大鼠单侧黑质内注入鱼藤酮建立帕金森损伤模型,治疗组同时腹部注射LA;采用免疫组化法观察黑质酪氨酸羟化酶(TH)的表达,ELISA法检测黑质TNF-α、IL-1β炎症介质含量变化。结果与对照组比较,损伤组TH阳性细胞率和灰度值显著降低(P<0.05),治疗组高于损伤组,低于对照组(P<0.05);损伤组TNF-α和IL-1β含量减少(P<0.05),治疗组增高,但仍低于对照组(P<0.05)。结论 LA对鱼藤酮损伤老年大鼠DA能神经元细胞具有神经保护功能。
Objective To investigate the effect of lipoic acid (LA) on the survival and inflammatory mediators of dopaminergic neurons in substantia nigra of Parkinson’s rats. Methods Healthy adult male SD30 rats were randomly divided into control group, injury group and treatment group. Parkinson’s injury model was established by injection of rotenone into unilateral substantia nigra of injured group and treatment group by stereotactic technique. At the same time, LA. The tyrosine hydroxylase (TH) expression in substantia nigra was observed by immunohistochemistry. The levels of TNF-α and IL-1β in the substantia nigra were detected by ELISA. Results Compared with the control group, the percentage of TH-positive cells in the injury group was significantly lower (P <0.05), and the level of TH-positive cells in the injury group was higher than that in the injury group (P <0.05). The levels of TNF-α and IL- Content decreased (P <0.05), the treatment group increased, but still lower than the control group (P <0.05). Conclusion LA has neuroprotective effect on DA neurons in rotenone-injured aged rats.
