目的探讨白三烯受体1(CYSLTR1)和白三烯C4合成酶(LTC4S)基因多态性与上海儿童哮喘及其严重程度之间的关系。方法 2006年9月至2009年8月本院儿科住院及门诊儿童哮喘患者72例,男39例,女33例,平均年龄(5.1±1.6)岁。根据哮喘发作程度分为间歇和轻度发作组44例、中重度发作组28例。健康对照组成人95例,男50例,女45例。应用连接酶检测反应(LDR)技术,对CYSLTR1的927T/C位点和LTC4S的444A/C位点基因多态性进行分析。结果①哮喘组和对照组927T/C位点基因型、等位基因频率比较(χ2=0.178,0.04,P(0.05)、444A/C位点基因型、等位基因频率比较(χ2=1.14,1.065,P(0.05)差异均无统计学意义。②927T/C和444A/C位点基因型频率在哮喘各组中的分布比较(χ2=0.927,1.138,P(0.05)差异均无统计学意义。结论在上海哮喘儿童中,CYSLTR1的927T/C位点和LTC4S的444A/C位点基因多态性可能不是其发病和严重程度的主要基因危险因素。
Objective To investigate the relationship between polymorphisms of leukotriene receptor 1 (CYSLTR1) and leukotriene C4 synthase (LTC4S) gene and the severity of asthma in Shanghai children. Methods From September 2006 to August 2009, 72 pediatric patients with pediatric asthma inpatients and outpatients, 39 males and 33 females, with an average age of 5.1 ± 1.6 years. According to the extent of asthma attacks were divided into intermittent and mild attack in 44 cases, moderate-severe attack in 28 cases. 95 healthy adults, 50 males and 45 females. The polymorphisms of CYSLTR1 927T / C locus and LTC4S 444A / C locus were analyzed by ligase detection reaction (LDR). Results ① The genotype and allele frequency of 927T / C locus in asthma group and control group were compared (χ2 = 0.178,0.04, P (0.05), 444A / C genotypes and allele frequencies (χ2 = 1.14, 1.065, P (0.05), respectively.②The genotype frequencies of 927T / C and 444A / C in asthmatic groups were not significantly different (χ2 = 0.927,1.138, P <0.05) .Conclusion Among the asthmatic children in Shanghai, the polymorphisms of 927T / C locus of CYSLTR1 and 444A / C locus of LTC4S may not be the major genetic risk factors of their onset and severity.
