目的:建立高效液相色谱蒸发光散射检测(HPLC-ELSD)法测定穿山龙中薯蓣皂苷元的含量,并与2005年版《中国药典》方法紫外检测器(DAD)测定的结果进行比较。方法:Agilent EclipseXDB-C18柱(4.6 mm×150 mm,5μm),流动相甲醇-水(95:5),柱温40℃;ELSD检测器漂移管温度为70℃,载气压力367.5 Pa,Gain7,Filter1;进样量10μL。结果:薯蓣皂苷元在2.60~20.8μg线性关系良好(r=0.999 94),平均回收率为97.3%,RSD 1.94%。结论:该方法简便,灵敏,准确性高,可作为穿山龙中薯蓣皂苷元含量测定的方法;与DAD相比,该法所得图谱基线平整,检测灵敏度高,样品分离度好,测得样品含量偏高。
OBJECTIVE: To establish a method for the determination of diosgenin in Chuan Shanlong by high performance liquid chromatography with evaporative light scattering detection (HPLC-ELSD) and compare with the results of the 2005 edition of “Chinese Pharmacopoeia” UV detector (DAD). Methods: Agilent EclipseXDB-C18 column (4.6 mm × 150 mm, 5 μm) was used. The mobile phase consisted of methanol and water (95: 5) with a column temperature of 40 ℃. The drift tube temperature was 70 ℃, the carrier gas pressure was 367.5 Pa, , Filter1; Injection volume 10μL. Results: The linear range of diosgenin was 2.60 ~ 20.8μg (r = 0.999 94). The average recovery was 97.3% and the RSD was 1.94%. Conclusion: The method is simple, sensitive and accurate, and can be used as a method for determination of diosgenin in Dangshan longan. Compared with DAD, the method has the advantages of flat baseline, high sensitivity and good sample separation high.
