目的:探讨益气化瘀方对神经再生修复过程中神经肌肉接头部纤维连接蛋白(fibronectin FN)的作用。方法:大鼠L5神经根受压后,分别予以益气化瘀方灌胃治疗10、30和60d后取比目鱼肌。以FN作为细胞外基质标记,α-bunga-rotoxin(α-BTX)萤光结合剂显示乙酰胆碱受体(acetylchline receptor,AChR),采用免疫组织化学方法结合激光共聚焦扫描显微(Confocal Iaser Scanning Microscopy system)技术,观察大鼠比目鱼肌神经肌肉接头部FN的表达,NI H图像分析技术定量测定FN与AChR的重叠面积。结果:大鼠L5神经根损伤后10d和30d,益气化瘀方组FN的表达及与乙酰胆碱受体(Acetylchline Receptor,AChR)的重叠面积均明显高于模型组,60d时,益气化瘀方组FN的表达较模型组显著回落,AChR的形态再构建明显优于模型组。结论:FN的表达与运动神经终板的神经再支配状况密切相关,益气化瘀方可促进FN表达,加快神经再生修复进程。
Objective: To investigate the effect of Yiqi Huayu Recipe on fibronectin FN of neuromuscular junction in the process of nerve regeneration. METHODS: Rat L5 nerve roots were compressed and then treated with Yiqi Huayu Recipe for 10, 30, and 60 days respectively to obtain soleus muscle. Using FN as an extracellular matrix marker, α-bunga-rotoxin (α-BTX) fluorescent binder shows acetylcholine receptor (AChR) using immunohistochemistry in combination with confocal Iaser Scanning Microscopy. (System) technique was used to observe the expression of FN in the neuromuscular junction of rat soleus muscle. NI H image analysis technique was used to quantitatively determine the overlapping area between FN and AChR. RESULTS: At 10 days and 30 days after L5 nerve root injury in rats, the expression of FN and the overlap area of acetylcholine receptor (AChR) were significantly higher in the Yiqi Huayu Recipe group than in the model group. At 60 days, the FN was increased. The expression of FN in the group was significantly lower than that in the model group, and the reconstitution of the AChR was significantly better than that of the model group. Conclusion: The expression of FN is closely related to the nerve innervation of motor nerve endplates. Yiqi Huayu Decoction can promote the expression of FN and accelerate the process of nerve regeneration.
