目的:通过比较飞蓬蛋白的不同提取方法,寻找一种简单、可行和有效的飞蓬蛋白提取方法。方法:采用现代提取植物蛋白的几种方法对飞蓬蛋白进行提取,并利用SDS-聚丙烯酰胺凝胶电泳法跟踪蛋白提取情况,同时依据Lowry法[1]所测定的飞蓬总蛋白含量,对提取方法进行优选。结果:硫酸铵沉降法所得电泳图谱条带最多,最清晰,Lowry法测其蛋白含量也最高(51.7%)。结论:硫酸铵沉降法得到的飞蓬蛋白含量及纯度较合理,方法更可行。
OBJECTIVE: To find out a simple, feasible and effective method for the extraction of Scrophularidium by comparing different extraction methods of Scrophulariaceae. Methods: Several methods of modern plant protein extraction were used to extract the extract of P. scoparia. SDS-polyacrylamide gel electrophoresis was used to trace the protein extraction. At the same time, the total protein content of P. scrofa was determined by Lowry’s method [1] Method is preferred. Results: The bands of electropherogram obtained by ammonium sulfate precipitation method were the most and the most clear. The content of protein was the highest (51.7%) by Lowry method. Conclusion: The content and purity of sclater canopy protein obtained by ammonium sulfate sedimentation method is more reasonable and the method is more feasible.
