目的观察巨噬细胞趋化抑制因子(MIF)及核转录因子κB/P65(NF-κB/P65)在单侧输尿管梗阻大鼠肾小管间质中的表达及1,25-(OH)2-D3对其的影响。方法采用单侧输尿管结扎手术建立单侧输尿管梗阻模型。将健康成年的雄性SD大鼠按随机数字表法分为3组:假手术组(n=10)、模型组(n=10)和VD组(n=10)。VD组采用6ng/(100 g·d)的1,25-(OH)2-D3干预治疗,假手术组和模型组仅给予等量生理盐水灌胃8周。观察3组大鼠第8周肾功能、肾组织病理学改变,免疫组化法检测单核/巨噬细胞表面特异性标志抗原(ED-1)、NF-κB/P65、MIF在肾组织中的表达。结果与假手术组相比,模型组血肌酐增高(P<0.01),肾间质面积/统计场面积增加(P<0.01),肾组织ED-1阳性细胞明显增加(P<0.01),肾组织MIF的表达显著升高(P<0.01),NF-κB/P65核阳性细胞明显增加(P<0.01)。与模型组比较,VD组大鼠肾组织病理学变化得到一定程度的改善,肾组织ED-1、MIF的表达及NF-κB/P65核阳性细胞明显减少(P<0.01)。结论单侧输尿管梗阻大鼠肾间质MIF的表达和NF-κB/P65的活性明显增加,1,25-(OH)2-D3可能通过下调MIF的表达,抑制NF-κB/P65的活性来减轻肾小管间质的炎症反应,改善肾脏纤维化。
Objective To observe the expression of macrophage chemotactic factor (MIF) and nuclear factor κB / P65 (NF-κB / P65) in the tubulointerstitium of rats with unilateral ureteral obstruction and the effects of 1,25- (OH) 2- D3 on its impact. Methods Unilateral ureteral obstruction was established by unilateral ureteral ligation. Healthy adult male Sprague-Dawley rats were randomly divided into 3 groups: sham operation group (n = 10), model group (n = 10) and VD group (n = 10). The VD group was treated with 6ng / (100 g · d) 1,25- (OH) 2-D3, and the sham-operation group and model group were given only the same amount of saline for 8 weeks. The changes of renal function and pathological changes of kidney in the third week were observed. The expression of monocyte / macrophage specific surface antigen (ED-1), NF-κB / P65 and MIF in renal tissue were detected by immunohistochemistry expression. Results Compared with the sham operation group, serum creatinine increased (P <0.01) and the area of renal interstitium increased (P <0.01), and ED-1 positive cells in renal tissue increased significantly (P <0.01) The expression of MIF was significantly increased (P <0.01), and the number of nuclear NF-κB / P65 positive cells was significantly increased (P <0.01). Compared with model group, the pathological changes of kidney in VD group were improved to a certain degree, the expression of ED-1 and MIF in renal tissue and the number of NF-κB / P65 nuclear positive cells were significantly decreased (P <0.01). Conclusion The expression of MIF and the activity of NF-κB / P65 in unilateral ureteral obstruction rats increased significantly. 1,25- (OH) 2-D3 may inhibit the activity of NF-κB / P65 by down-regulating the expression of MIF Reduce tubulointerstitial inflammatory response and improve renal fibrosis.
