308nm准分子光对白癜风豚鼠模型Treg和Th17细胞的影响

目的:通过白癜风豚鼠模型,观察308nm准分子光的疗效及对Treg、Thl7细胞的影响,探讨紫外光疗机制中的免疫学通路。方法:通过文件检索使用化学脱色法制备白癜风豚鼠模型并筛选造模成功的小鼠作为实验对象,将小鼠分为实验组和对照组,另选取相同数量的健康小鼠作为空白组,对实验组进行308nm准分子光的干预治疗,造模成功的对照组以及健康小鼠的空白组均不予以处理,10周后分别从肉眼复色程度、组织病理及免疫组化结果中的改变、mRNA以及蛋白表达的水平观察IL-17细胞和Treg细胞在三组之间的实验过程中的变化趋势。结果:308准分子光对白癜风豚鼠模型达到治疗效果,经过10周治疗后,5只豚鼠达到完全复色。在组织病理中,白癜风模型组豚鼠皮损中表皮黑素细胞减少,伴随真皮淋巴细胞浸润。治疗后,从组织学水平观察到,黑素细胞的增多,以及淋巴细胞浸润减少。免疫组织化学染色对皮损中IL-17标记可见,造模成功组中真皮层可见阳性结果的出现,此模型可用于本课题的研究。通过对皮损中IL-17及Foxp3的mRNA及蛋白表达水平定量检测发现,模型组IL-17表达升高,Foxp3表达降低,治疗后IL-17表达水平降低,Foxp3表达水平升高。结论:308nm准分子光疗可以通过降低豚鼠白癜风皮损中Th17细胞水平,升高Treg细胞水平调节皮损中Th17和Treg细胞的平衡紊乱。 OBJECTIVE: To observe the curative effect of 308nm excimer light and its effect on Treg and Thl7 cells in Vitiligo guinea pig model and to explore the immunological pathway in UV phototherapy. Methods: Vitiligo guinea pig model was prepared by chemical decoloration through file retrieval, and the successful model mice were screened. The mice were divided into experimental group and control group, and the same number of healthy mice were selected as blank group. Group 308nm excimer light intervention treatment, the successful control group and healthy mice blank group were not treated, 10 weeks after the eye from the degree of re-staining, histopathology and immunohistochemical results, mRNA And the level of protein expression were observed IL-17 cells and Treg cells in the three groups during the experiment trend. Results: The 308 excimer light reached the therapeutic effect on the model of guinea pig with vitiligo. After 10 weeks of treatment, the five guinea pigs achieved complete re-colorization. In histopathology, there was a decrease in epidermal melanocytes in guinea pigs with vitiligo, accompanied by infiltration of dermal lymphocytes. After treatment, it was observed from the histological level that melanocytes increased and lymphocyte infiltration decreased. Immunohistochemical staining of IL-17 markers in the lesion can be seen in the successful model group dermal layer visible positive results, this model can be used for the study of this topic. Quantitative detection of IL-17 and Foxp3 mRNA and protein expression in the lesion showed that the expression of IL-17 in the model group was increased, the expression of Foxp3 was decreased, the expression of IL-17 was decreased and the expression of Foxp3 was increased in the model group. CONCLUSION: 308 nm excimer phototherapy can regulate Th17 cell level in guinea pig vitiligo lesions and increase Treg cell level to regulate the balance disorder of Th17 and Treg cells.
