本研究是在一九六六年和一九六七年一~三月份进行的。试验用鱼取自琦玉县水产试验场熊谷养鳟试验池养成的3~4令的雌虹鳟。方法是使排卵后的卵,长时间地停留在母体内,从排卵时算起,以后每隔3天(67年)和5天(66年)挤出部分卵,观察卵在过熟过程中的外部形态、组织构造以及遇水被激活后的形态变化等。普通组织学检查以Bauin氏液或20%福尔马林液为固定剂,石蜡包埋作成切片,以Mayer氏酸性铝苏木精—伊红、Heidenheirn氏azan染色。组织化学方面以高碘酸—schiff(PAS)法勘定细胞中皮质泡,用苏丹Ⅲ或苏丹黑B染色的冰冻切片勘定细胞中的油球。
This study was conducted in 1966 and January-March 1967. The test fish was collected from 3 to 4 female rainbow trout formed from the Xionggu grazing test pond at the Saitama Aquatic Experimental Field. Method is to make ovulation eggs, stay in the mother for a long time, from the time of ovulation, after every 3 days (67 years) and 5 days (66 years) out of some of the eggs to observe the eggs in the process of over-ripening The external shape, organizational structure, and morphological changes after water is activated. Histochemistry: The intracellular cortical vesicles were identified by period-s-chiff (PAS) and frozen sections of cells stained with either Sudan III or Sudan Black B were used to assess the oil globules.
