通过流式细胞仪的方法,报道了凝集素美洲商陆(PWM)对人单核细胞和白血病细胞表面的HLA-DR分子表达的影响。实验结果显示:PWM可以直接与单核细胞和白血病U937细胞及Raji细胞相结合,而且这3种细胞均表达HLA-DR分子。经PWM处理24h后,正常人单核细胞HLA-DR分子的表达不受影响,而U937细胞和Raji细胞上的HLA-DR分子均被下调,而且这种下调作用与PWM的剂量有关。其他凝集素,如PHA、WGA和内毒素LPS对白血病细胞HLA-DR分子的表达无影响。结果表明,通过凝集素PWM的处理可以改变肿瘤细胞表面MHCⅡ类分子的表达,从而为肿瘤免疫治疗提供新的途径。
The effect of lectin, pokeweed, on the expression of HLA-DR molecules on the surface of human monocytes and leukemia cells was reported by flow cytometry. Experimental results show that: PWM can be directly associated with monocytic and leukemic U937 cells and Raji cells, and all three cells express HLA-DR molecules. After treated with PWM for 24 h, the expression of HLA-DR in normal human monocytes was not affected, while the HLA-DR molecules in U937 cells and Raji cells were downregulated, and this down-regulation was related to the dose of PWM. Other lectins, such as PHA, WGA and LPS, had no effect on HLA-DR expression in leukemia cells. The results showed that the treatment of lectin PWM can change the expression of MHC class II molecules on the surface of tumor cells, thus providing a new way for tumor immunotherapy.
