目的:探讨凉血活血方(LiangXue Huo Xue Fang)对肝星状细胞LX-2增殖和凋亡的影响。方法:体外培养肝星状细胞LX-2,分别将凉血活血方(浓度为1.125,2.228,3.31,4.37,5.41 mg·mL-1),作用于LX-2细胞后24 h,48 h用MTT比色法检测细胞的增殖情况,选取合适的药物浓度(4,8,12,16 mg·mL-1),用流式细胞术检测加药24 h后细胞的凋亡率。结果:1MTT法显示不同浓度凉血活血方作用LX-2细胞24 h,48 h后,除24 h,1.125 mg·mL-1和2.228 mg·mL-1外,其余组增殖抑制率均明显高于正常对照组(P<0.05)。2流式细胞术检测显示,凉血活血方浓度为4,8,12,16 mg·mL-1)时,可明显促进LX-2细胞凋亡。结论:一定剂量的凉血活血方能抑制LX-2细胞的增殖,可能与其促进细胞凋亡有关。
Objective: To investigate the effect of Liangxue Huo Xue Fang on the proliferation and apoptosis of hepatic stellate cells LX-2. METHODS: Rat hepatic stellate cells LX-2 were cultured in vitro and the blood-cooling and blood-activating agents (concentration 1.125, 2.228, 3.31, 4.37, 5.41 mg·mL-1) were applied to LX-2 cells 24 h and 48 h later. MTT colorimetric assay was used to detect the proliferation of the cells. The appropriate drug concentrations (4, 8, 12, 16 mg·mL-1) were selected. Flow cytometry was used to detect the apoptosis rate of the cells after dosing for 24 h. RESULTS: 1 MTT assay showed that LX-2 cells were treated with different concentrations of Liangxue Huoxue Prescription for 24 h. After 48 h, except for 24 h, 1.125 mg·mL-1 and 2.228 mg·mL-1, the proliferation inhibition rates of the other groups were significantly higher. In the normal control group (P <0.05). 2 Flow cytometry showed that when the concentration of Liangxue Huoxue Decoction was 4, 8, 12, 16 mg·mL-1), it could significantly promote LX-2 cell apoptosis. Conclusion: A certain dose of Liangxue Huoxue can inhibit the proliferation of LX-2 cells, which may be related to its promotion of cell apoptosis.
