目的探讨山芝麻(RH)水提取物对百草枯(PQ)致大鼠肺纤维化Sonic hedgehog(SHH)信号途径Smo和Gli1蛋白表达的影响,为山芝麻水提取物治疗PQ致SHH提供理论依据。方法 60只雄性大鼠随机分为PQ组、生理盐水对照组(NS组)、PQ组+RH水提取物组(PQ+RH组),观察3组大鼠肺组织外观及病理学改变;运用免疫组化法及斑点杂交(Western-blot)法检测肺组织SHH信号途径的Smo和Gli1的表达。结果 PQ+RH组大鼠肺组织较PQ组纤维化程度减轻。检测Smo及Gli1在NS组无表达或弱表达;PQ组表达明显增强;PQ+RH组Smo及Gli1表达明显减弱;组间比较,差异有统计学意义(P<0.05)。结论 RH水提取物显著减低SHH信号途径Smo和Gli1蛋白表达,对SHH有一定的干预作用。
Objective To investigate the effect of aqueous extract of sesame (RH) on the expression of Smo and Gli1 protein in lung tissue of Sonic hedgehog (SHH) induced by paraquat (PQ) in rats and to provide a theoretical basis for the treatment of Pseudoscedit . Methods Sixty male rats were randomly divided into PQ group, saline control group (NS group) and PQ group + RH water extract group (PQ + RH group). The lung tissue appearance and pathological changes of the three groups were observed. The expression of Smo and Gli1 in SHH signal path of lung tissue were detected by immunohistochemistry and Western-blot. Results Compared with PQ group, the degree of fibrosis in lung tissue of PQ + RH group was relieved. The expression of Smo and Gli1 was not detected in NS group. The expression of Smo and Gli1 was significantly decreased in PQ + RH group. The difference was statistically significant (P <0.05). Conclusion RH extract can significantly reduce the expression of Smo and Gli1 protein in SHH signaling pathway, which may have some effects on SHH.
