不同生长基质对神经干细胞分化为类毛细胞影响的实验研究

目的探讨新生豚鼠海马神经干细胞在人工外淋巴液中分化为类毛细胞的最佳生长条件。方法将原代培养的新生豚鼠海马来源的神经干细胞进行体外培养(15%人工外淋巴液+DMEM/F12),分别接种于包被多聚赖氨酸和鼠尾胶原的盖玻片上使其分化,3周后对分化的细胞进行免疫荧光检测,分别计数产生表达毛细胞标记蛋白MysionⅦa阳性细胞的比例。结果神经干细胞在接种鼠尾胶原的盖玻片上生长,分化为MysionⅦa阳性细胞的比例为13.6%。但在接种多聚赖氨酸的盖玻片上生长,MysionⅦa阳性细胞的比例仅为1.9%。结论鼠尾胶原是适合新生豚鼠海马神经干细胞在人工外淋巴液中分化为表达毛细胞特异抗体细胞的生长基质。 Objective To investigate the optimal growth conditions of neonatal guinea pig hippocampal neural stem cells differentiated into hair cells in artificial perilymph. Methods Primary cultured neonatal guinea pig hippocampus-derived neural stem cells were cultured in vitro (15% artificial perilymph + DMEM / F12) and inoculated on coverslips coated with polylysine and rat tail collagen to differentiate After 3 weeks, the differentiated cells were detected by immunofluorescence, and the percentage of cells expressing the hair cell marker protein MysionⅦa was counted. Results Neural stem cells grew on coverslips inoculated with rat tail collagen and the percentage of differentiated into Mysion Ⅶ a positive cells was 13.6%. However, when grown on polylysine-coated coverslips, the percentage of Mysion VIIa-positive cells was only 1.9%. Conclusion The rat tail collagen is suitable for the differentiation of hippocampus neural stem cells of neonatal guinea pigs into the growth media that express the hair cell specific antibody in the artificial perilymph.
